What about Intact Protein?
It is difficult to remove intact proteins from gels. We list here some procedures that MIGHT be useful for removing intact protein.
You can follow the listed protocols without using trypsin. Acetonitrile will be able to extract some protein off the gel, then the extract can be speed-vacuumed and run by MALDI-MS. However, the best way to get the intact protein out of the gel will be electro-elution.
Recent research found that by shaking highly acidic/basic buffer with the gel, the proteins can be removed from the gel. For more information, a paper by Steven L. Cohen and Brian T. Chait is linked here.
